Un messaggio criptico proveniente dal suo passato manda James Bond (Daniel Craig) in missione prima a Città del Messico e poi a Roma, dove incontra Lucia Sciarra (Monica Bellucci), la bella e proibita vedova di un infame criminale. Infiltrandosi in un incontro segreto, Bond scopre così l'esistenza di una pericolosa organizzazione nota come SPECTRE. Nel frattempo a Londra Max Denbigh (Andrew Scott), il nuovo capo del centro per la sicurezza nazionale, mette in discussione l'operato di Bond e contesta l'importanza del MI6, guidato da M (Ralph Fiennes). Bond, inoltre, arruola segretamente Moneypenny (Naomie Harris) e Q (Ben Whishaw) per essere aiutato a stanare chi e cosa sia dietro a SPECTRE. Una ricerca che passa per Madeleine Swann (Léa Seydoux), la figlia del vecchio nemico White (Jesper Christensen), che potrebbe avere la chiave per districare la rete dell'organizzazione.
