Paterson guida gli autobus nella città del New Jersey che porta il suo stesso nome. Ogni giorno rispetta una sorta di routine oramai collaudata: attraversa il solito percorso, osserva la città alla deriva dal suo parabrezza, ascolta frammenti delle conversazioni della gente intorno a lui, scrive poesie, va a fare passeggiate con il suo cane, si ferma in un bar e beve una birra. Poi va a casa dalla moglie Laura, il cui mondo è invece in continuo cambiamento, fatto di sogni e progetti diversi e ispirati. I due si amano: mentre Paterson sostiene le ambizioni di lei, Laura apprezza in segreto il suo talento per la poesia. Nel corso di una settimana, si osservano i loro trionfi e le loro sconfitte, così come quelle dell'intera cittadina di Paterson.
